Overview

Dynein light chain Tctex-type 1 (DYNLT1) is a protein encoded by the DYNLT1 gene in humans, functioning as a non-catalytic accessory component of the cytoplasmic dynein motor complex. It is responsible for linking dynein to cargos and adapter proteins for intracellular retrograde motility along microtubules. DYNLT1 plays additional roles in regulating neuronal morphogenesis independently of dynein, and it interacts with viral proteins to facilitate infection processes. Its expression and interactions influence pathways such as immune system function, neurogenesis, and mitotic spindle orientation. Dysregulation of DYNLT1 or its interactions has been associated with various diseases, including cancer and certain infections. The structural characterization of DYNLT1 reveals it forms homodimers that bind dynein intermediate chains and adapter proteins, operating as both a molecular clamp and a microtubule-cargo adapter.
