Overview

Dynein light chain Tctex-type 3 (DYNLT3) is a non-catalytic accessory component of the cytoplasmic dynein 1 motor protein complex, mainly involved in intracellular retrograde transport of vesicles and organelles along microtubules, and in mitotic progression by binding cargoes such as spindle checkpoint protein BUB3[1][3]. It can also operate independently of the dynein complex as a transcriptional modulator, notably regulating Bcl-2 via interaction with SATB1[1][3]. Differentially expressed across tissues, DYNLT3 expression has been linked to enhanced proliferation, migration, and invasion in tumor models such as ovarian cancer, and its high expression correlates with poor prognosis in this context[5]. There are no approved drugs or known direct ligands for DYNLT3, and while not a classic therapeutic target such as an enzyme or receptor, it represents a potential biomarker and contributor to cancer biology through modulation of cell cycle and cytoskeletal dynamics[5].
