Overview

Dynein regulatory complex subunit 1 (DRC1) is a core component of the nexin-dynein regulatory complex (N-DRC), a large, evolutionarily conserved scaffold that regulates the assembly and activity of axonemal dynein motors within motile cilia and flagella[1][2][3][4][5][7]. The N-DRC spans between microtubule doublets within the axoneme and regulates the sliding motion that produces ciliary and flagellar beating[1][2][5]. DRC1 is critical for the assembly and stability of the N-DRC, ensuring proper ciliary motility and structural integrity of the axoneme[3][5]. Mutations in DRC1 cause human diseases such as primary ciliary dyskinesia and spermatogenic failure, both characterized by impaired motile cilia or flagella function[3][4]. DRC1 forms a central scaffold with other N-DRC subunits and coiled-coil domain-containing proteins, linking and regulating dynein arms and supporting the correct positioning and coordination required for effective ciliary movement[1][2][6]. Additional notes: DRC1 is not a classical drug target (not a receptor, enzyme, transporter, etc.), so there are no direct pharmacologic modulators or established biomarkers for it[3][7].
