Overview

Dynein regulatory complex subunit 3 is a protein component of the nexin-dynein regulatory complex (N-DRC), a conserved multi-protein assembly essential for the regulation of ciliary and flagellar motility in eukaryotic cells[2][3]. The N-DRC forms a cross-bridge between neighboring outer doublet microtubules in the axoneme and regulates the activity of axonemal dynein motors, thereby controlling the sliding and bending motions required for effective ciliary and flagellar beating[1][2]. DRC3 is classified as a “functional subunit” of the N-DRC and acts as an adapter required for the proper assembly of other functional components during axoneme formation, especially in sperm flagella maturation and motility[2][3]. Loss or dysfunction of DRC3 disrupts the structural organization and regulatory function of the N-DRC, leading to impaired ciliary beating and can contribute to human disorders such as primary ciliary dyskinesia[2][3]. There are currently no drugs known to target DRC3 directly, and its status as a druggable therapeutic target has not been established.
