Overview

Dynein regulatory complex subunit 5 (DRC5) is a component of the nexin-dynein regulatory complex (N-DRC), a multi-protein complex located in the axoneme of motile cilia and flagella. DRC5 is a leucine-rich subunit that interacts with other scaffolding proteins (DRC1/2, DRC4A/B) within the linker region of the N-DRC[1][4]. The N-DRC itself functions as an essential regulator and structural linker, controlling dynein motor activity and thus the waveform and motility of cilia and flagella[1][3][4][5]. DRC5 and its associated subunits play critical roles in the stability and regulatory function of the N-DRC; loss or mutation of DRC5 disrupts ciliary motility but does not completely destabilize complex assembly[3][4][5]. The DRC5 gene is also known as TCTE1, and the corresponding protein has been studied in organisms including Chlamydomonas and mammals for its role in sperm flagellar motility and respiratory cilia function. Currently, DRC5 is not considered a direct therapeutic target and no known drugs are reported to interact with it[1][3][4][5].
