Dynein regulatory complex subunit 8 (DRC8) is a component of the highly conserved nexin–dynein regulatory complex (N-DRC) found in motile cilia and flagella of eukaryotic cells[2][4]. This complex acts as a linker between neighboring microtubule doublets, stabilizes the axonemal core structure, and plays a central regulatory role in ciliary motility by coordinating dynein motor activity[1][2][4]. DRC8 contains EF-hand calcium-binding domains, implicating it in calcium-mediated signaling relevant to ciliary motility[2]. DRC8 and DRC11 form a subcomplex localized to the proximal lobe of the N-DRC linker region, crucial for the structural interface between the complex and the adjacent B-tubule[2][4]. While DRC8 is essential for normal flagellar function, mutations usually result in minor defects in motility, and it has not been directly implicated as a drug target or biomarker for disease[2][4].
