Overview

E1A binding protein p400 (EP400) is a large, multi-domain ATP-dependent chromatin remodeling factor and a core component of the human TIP60–EP400 complex, which regulates chromatin structure through both histone exchange and histone acetylation[1][2][3]. It acts as a scaffold essential for assembling this multi-protein complex and is directly involved in DNA repair via homologous recombination, transcriptional activation of genes (notably by acetylating histones H2A/H4), and cell cycle regulation[1][2][3]. EP400 binds and modulates chromatin at sites of DNA double-strand breaks, facilitates the recruitment of DNA repair machinery, and is implicated in regulating key pathways governing apoptosis and cell fate decisions—particularly via interactions with oncogenic and tumor suppressor factors such as p53 and Myc[1][3]. Dysregulation or altered expression of EP400 has been associated with several cancers. While under investigation for its role in tumorigenesis and as a potential biomarker, there are currently no approved drugs directly targeting EP400 or confirmed disease biomarkers based on EP400 modulation[1][3].
