Overview

The E26 transformation-specific (ETS) transcription factor family is a group of approximately 28 human proteins characterized by a conserved 84-amino-acid DNA-binding domain known as the ETS domain, which recognizes purine-rich motifs, typically GGAA/T, in the promoters and enhancers of target genes. The family controls diverse cellular processes: development, differentiation, proliferation, apoptosis, angiogenesis, hematopoiesis, and inflammation. Members are divided into subfamilies (such as ETS, ERG, ELK, ELF, and PEA3) and include tumor-associated factors like ETS-1, ERG, and ETV6 as well as tissue-specific proteins like ESE-1/ELF3. Dysregulation or mutation in family members is implicated in cancer (e.g., ETV5 in colorectal and breast cancer; ETS-1 in vascular neointima and cancer), cardiovascular, and inflammatory diseases. Therapeutic targeting is challenging due to redundancy and broad functions but remains of substantial interest in oncology and vascular biology.
