Overview

An E2F-responsive promoter is a specific region of DNA containing E2F-binding sites, which serve as a platform for E2F family transcription factors to regulate the expression of nearby genes. E2F-responsive promoters are critical for controlling expression of genes involved in cell cycle progression, DNA replication, apoptosis, and proliferation. The activity of these promoters is tightly regulated by activating or repressing E2F proteins, often in cooperation with pocket proteins (such as pRB, p107, p130); their binding dynamically changes with cell cycle phase, contributing to either repression (in quiescence/early G1) or activation (late G1/S phase). Aberrant regulation at E2F-responsive promoters is associated with uncontrolled cell growth and cancer. However, these promoters are not direct drug targets; rather, their regulatory components—namely the E2F proteins—are the relevant biological targets.
