E2F transcription factor 2 (E2F2) is a key regulator of cell cycle, proliferation, and apoptosis that binds to DNA at specific promoter regions to modulate gene expression. Recent studies have demonstrated that E2F2 directly binds to the promoter of MyD88, a critical adaptor protein in innate immune signaling, and upregulates its expression, activating downstream PI3K/AKT/NF-κB signaling implicated in inflammation and cancer[5][3]. This interaction contributes to aggressive cellular phenotypes, such as proliferation, invasion, and cytokine production, especially in rheumatoid arthritis and cancer. Although the term "E2F2-MyD88 promoter complex" describes a relevant regulatory event, it is not a standardized molecular target and should be interpreted as the binding of E2F2 to the MyD88 promoter to control MyD88-dependent signaling pathways.
Drugs may act by inhibiting E2F2's gene expression or DNA binding activity, thereby affecting cell cycle progression, proliferation, or immune signaling (e.g., via PI3K/AKT/NF-κB pathways downstream of MyD88). ST2825 targets MyD88-dependent pathways, indirectly affecting E2F2-regulated gene expression.
