Overview

E2F transcription factor 5 (E2F5) is a member of the E2F family of transcription factors that plays a pivotal role in controlling the cell cycle, particularly during the transition from G0/G1 to S phase (UniProt Q15329). Unlike the activator E2Fs, E2F5 primarily acts as a transcriptional repressor by associating with pocket proteins like p130 to silence genes necessary for DNA replication (NCBI Gene ID: 1875). In many human cancers, such as ovarian, breast, and prostate cancer, E2F5 is significantly upregulated, where it promotes tumor cell proliferation, survival, and metastasis (PMID: 29328343). Because E2F5 lacks a traditional druggable pocket for small molecules, therapeutic interventions often target its mRNA transcript using RNA interference (RNAi) or antisense technologies to reduce protein levels (PMID: 21833015). Research indicates that silencing E2F5 mRNA can inhibit tumor growth and sensitize cancer cells to chemotherapy, making it a high-interest target for nucleic acid-based therapeutics (PMID: 25670303). Additionally, E2F5 expression levels are being investigated as diagnostic and prognostic biomarkers in various solid tumors.
