Overview

E2F transcription factor 8 (E2F8) is an atypical member of the E2F family of transcription factors involved in the regulation of cell cycle and transcriptional repression or activation of target genes. E2F8 is implicated in cell cycle progression, especially the G1/S transition, and can act as both an oncogene and tumor suppressor in different contexts. In basal-like breast cancer, E2F8 is typically upregulated and associated with poor prognosis, increased cell proliferation, and immune cell infiltration. It regulates genes involved in mitosis and can interact with other cancer-related proteins such as MYC, TP53, and BRCA1. Knockdown of E2F8 inhibits cancer cell migration, invasion, and enhances sensitivity to chemotherapy (docetaxel). Its high expression may also predict better responses to immune checkpoint inhibitor therapies[1].

For docetaxel: Enhanced cell cycle inhibition and reduced proliferation upon E2F8 downregulation. For immune checkpoint inhibitors: Potentially increased efficacy in tumors with high E2F8 expression due to greater T-cell infiltration.
