Overview

IDOL (Inducible Degrader Of the LDL receptor) is an E3 ubiquitin ligase that plays a central role in the post-translational regulation of lipid metabolism and neuronal function. It is encoded by the *MYLIP* gene and is best known for targeting members of the low-density lipoprotein receptor (LDLR) family for degradation, thereby modulating cholesterol uptake and homeostasis. Expression of IDOL is directly regulated by liver X receptors (LXRs), which are nuclear receptors activated in response to increased cellular sterol levels.

Mechanism of action

IDOL mediates ubiquitination and subsequent lysosomal degradation of LDLR, VLDLR, and ApoER2, thereby reducing cholesterol uptake.
