E3 ubiquitin-protein ligase DTX1, also known as Deltex1, is a RING-type E3 ubiquitin ligase and a member of the Deltex family (UniProt, GeneCards). It plays a critical role in modulating the Notch signaling pathway by mediating the ubiquitination and subsequent degradation of various substrates, such as the Notch intracellular domain (NICD), MEKK1, and c-FLIP (Frontiers in Cell and Developmental Biology, MDPI). Through these interactions, DTX1 influences essential biological processes including cell-fate determination, T cell and B cell development, neurogenesis, and apoptosis (UniProt, NIH). In disease contexts, DTX1 is implicated in several cancers, such as diffuse large B-cell lymphoma (DLBCL) and gastric cancer, where it can function as either a tumor suppressor or an oncogene depending on the cellular environment (ASH Publications, NIH). It has also been identified as a potential therapeutic target for viral infections like Hepatitis B and for neuroinflammatory conditions following traumatic brain injury (NIH). While specific DTX1-targeted drugs are not yet in clinical use, compounds like gambogic acid and geldanamycin have been shown to modulate its expression, and it is considered a candidate for novel therapeutic strategies such as PROTACs (NIH, Open Targets).
