Overview

E3 ubiquitin-protein ligase RNF103 is an enzyme encoded by the RNF103 gene in humans. It contains a RING-H2 finger motif, characteristic of the RING family of E3 ubiquitin ligases, and is involved in the transfer of ubiquitin from E2 conjugating enzymes to specific substrate proteins, thereby targeting them for degradation via the ubiquitin–proteasome pathway. RNF103 is highly expressed in the normal cerebellum, with low expression in the cerebral cortex. In rodent models, the gene’s expression in the brain is inducible by antidepressant treatments and electroconvulsive therapy, suggesting a possible role in neuropsychiatric responses. The protein has key molecular functions in endoplasmic reticulum-associated degradation (ERAD) and broader roles in maintaining protein quality control within cells[1][2][4][5][10]. RNF103 is associated with diseases such as neuroschistosomiasis and epidermolysis bullosa acquisita, but its direct disease mechanisms are not fully established. No drugs that specifically interact with RNF103 are currently known, but modulation of its activity or expression may represent a future therapeutic strategy for conditions linked to ER stress or protein misfolding[1][4][8].
