Overview

E3 ubiquitin-protein ligase RNF13 (abbreviated as RNF13) is a transmembrane RING-type E3 ligase that localizes mainly to the endoplasmic reticulum (ER) and Golgi apparatus. As a member of the PA-TM-RING protein family, RNF13 contains an N-terminal protease-associated (PA) domain, a transmembrane region, and a C-terminal RING zinc finger domain. RNF13 facilitates the transfer of ubiquitin to specific substrate proteins, regulating various processes such as ER stress response, apoptosis (particularly through activation of the IRE1α-TRAF2-JNK signaling axis), immune modulation, lipid metabolism, and neuronal development. Dysregulation of RNF13 expression or function is implicated in several disease contexts, including pancreatic cancer, neurodevelopmental disorders (such as developmental and epileptic encephalopathy 73), and metabolic liver diseases. While RNF13 itself has not been directly targeted by approved drugs or clinical inhibitors, its emerging roles as a disease mediator make it a potential therapeutic target of interest.
