Overview

E3 ubiquitin-protein ligase RNF14 is a RING-type zinc finger protein encoded by the RNF14 gene in humans[1][4]. It functions primarily as an E3 ligase, mediating ubiquitination and subsequent proteasomal degradation of nuclear and translation-related proteins, especially in response to ribosomal stalling and cellular stress[2][4][5]. RNF14 acts as a transcriptional coactivator for the androgen receptor, stimulating androgen-responsive gene expression in prostate tissue, and as a co-regulator for TCF/LEF transcription factors, thereby potentiating Wnt/beta-catenin-driven gene transcription essential for colon cancer cell survival[1][4][8]. The protein also has a key role in regulating mitochondrial gene expression and immune-related gene networks, particularly in muscle cells, influencing the interrelationship between energy metabolism and inflammation[3][7]. Alternative splicing yields several isoforms with distinct regulatory capabilities[4][3]. RNF14 is considered a therapeutic target due to its involvement in cancer and cellular stress adaptation, although no direct drugs currently target it in clinical use.
