E3 ubiquitin-protein ligase RNF183 (RNF183) is a membrane-associated RING finger E3 ubiquitin ligase localized primarily to the endoplasmic reticulum. It catalyzes the attachment of ubiquitin moieties onto specific substrate proteins, leading to their proteasomal degradation. RNF183 is upregulated in response to prolonged endoplasmic reticulum stress, where it promotes apoptosis by targeting the antiapoptotic protein Bcl-xL for degradation. Additionally, RNF183 is highly expressed in the intestinal epithelium during inflammatory bowel disease and drives inflammation by ubiquitinating and degrading IκBα, leading to increased NF-κB signaling. Its expression is negatively regulated by miR-7, highlighting an epigenetic regulatory axis central to its proapoptotic and proinflammatory roles. RNF183’s functional significance spans ER stress, apoptosis, and chronic inflammation, positioning it as a promising, though still emerging, disease-relevant target.
