Overview

E3 ubiquitin-protein ligase RNF20 is a RING finger E3 ligase that primarily forms a complex with RNF40 to monoubiquitinate histone H2B at lysine 120. This modification serves as a key epigenetic marker that facilitates gene transcription and is prerequisite for further histone methylation at H3K4 and H3K79. RNF20 plays critical roles in regulating chromatin structure, DNA repair, and tumor suppression by activating p53 as well as controlling the expression of multiple histone and non-histone genes. It acts as a tumor suppressor but can also repress certain protooncogenes. RNF20 is implicated in maintaining genomic stability, modulating metabolic pathways in adipose tissue, and has been associated with various diseases including cancer, metabolic syndromes, and certain viral infections. No drugs are directly known to target RNF20 at this time, but it is a potential therapeutic target for diseases related to epigenetic dysregulation and cancer[1][2][4][5][7].
