E3 ubiquitin-protein ligase RNF212B is a RING finger domain-containing enzyme essential for crossover designation and maturation during meiosis I in mammals[1][2][3]. RNF212B acts as a ubiquitin E3 ligase, collaborating with paralogs such as RNF212 to ensure accurate formation of crossovers and proper segregation of homologous chromosomes. Its loss leads to defects in meiosis, primarily causing loss of crossovers and aberrant chromosome segregation, which can result in infertility[1][2]. RNF212B forms foci on chromosome axes in a synapsis-dependent but double-strand break (DSB)-independent manner, and colocalizes with major recombination factors such as HEI10, CNTD1, and MLH1[1][2]. Although not currently established as a therapeutic target or druggable target in disease, pathogenic variants have been implicated in reproductive disorders in both mice and humans[1]. There are no reports of drugs targeting RNF212B, nor is it currently regarded as a therapeutic target in clinical development. It is primarily classified as a meiosis-essential enzyme, with its main relevance to fertility, rather than to cancer, infection, or other major common diseases[1][2][3].
