Overview

E3 ubiquitin-protein ligase TRIM68 is a member of the tripartite motif-containing (TRIM) protein family, characterized by the presence of a RING finger domain, B-box motif, and a coiled-coil region[1][5][8]. TRIM68 acts as an E3 ubiquitin ligase, catalyzing the transfer of ubiquitin to substrate proteins, thereby regulating their stability and activity[1][2][5]. It serves as a coactivator of nuclear receptors, most notably the androgen receptor, facilitating transcriptional activation in a ubiquitin-dependent manner[1][2][4]. TRIM68 is highly expressed in prostate tissue and is upregulated in androgen-dependent prostate cancer cells, suggesting a role in cancer biology and hormone signaling[3]. Additionally, TRIM68 contributes to regulation of the innate immune response by negatively controlling the production of type I interferons and possibly modulating antiviral and inflammatory signaling pathways[5]. Alternative splicing results in multiple transcript variants encoding different isoforms[1]. Diseases associated with TRIM68 include prostate cancer, lupus erythematosus, and cataract[1]. Note: No small molecule drugs or antibodies are currently documented as direct modulators of TRIM68 in public databases or literature. Its clinical utility as a direct therapeutic target requires more research.
