E3 ubiquitin-protein ligase ZNRF4 (ZNRF4) is a member of the RING finger protein family characterized by a cysteine-rich zinc finger domain that coordinates two zinc ions, enabling its function as an E3 ligase in the ubiquitin-proteasome system[1][3][5]. ZNRF4 mediates the transfer of ubiquitin from E2 conjugating enzymes to specific substrate proteins, directing them for proteasomal degradation. It acts as a negative regulator of NOD2 signaling by ubiquitinating and promoting the degradation of RIPK2, which results in attenuation of innate immune responses[6]. ZNRF4 is implicated in immune tolerance and may contribute to the control of inflammation. No direct drug modulators or clinical biomarkers are currently established for ZNRF4, but alterations in its activity may influence immune-related disorders[6].
