Overview

The E5 oncogene protein is a small, hydrophobic transmembrane protein encoded by papillomaviruses. It promotes cell transformation, modulates growth factor receptor signaling (PDGFβR, EGFR), and impairs immune recognition. While critical in early stages of viral infection and tumorigenesis, it is often lost in advanced cancers, potentially limiting its therapeutic utility.

Mechanism of action

Modulates signal transduction pathways through interactions with key cellular proteins involved in growth regulation. Activates PDGFβR (BPV1) or enhances EGFR signaling by inhibiting its degradation (HPV16). Downregulates MHC class I expression.
