The E7 protein of human papillomavirus type 16 is a multifunctional oncoprotein essential for the virus-induced transformation of host cells. It is a small, nuclear and cytoplasmic phosphoprotein (about 98 amino acids), with three conserved regions: CR1 and CR2 (N-terminus, homologous to adenovirus E1A and SV40 large T antigen) and a C-terminal zinc-binding domain. E7’s main biological activity is binding to and inactivating the retinoblastoma tumor suppressor protein (pRb), resulting in release of E2F transcription factors and driving the host cell into S-phase, thus supporting viral replication and predisposing cells to malignant transformation. E7 lacks intrinsic enzymatic or DNA-binding activity and works by hijacking and degrading host regulatory proteins, cooperating with E6 (another HPV oncoprotein) to evade cell death and subvert cell cycle, contributing to the development and maintenance of HPV-associated cancers. The protein is considered a prime therapeutic target for vaccines and immunotherapies, with ongoing research aimed at developing medicines that disrupt its function. E7 mRNA or protein are used as diagnostic/prognostic biomarkers in HPV-driven malignancies, but safety and druggability challenges remain due to its viral origin, intracellular localization, and essential role in cell survival in HPV-transformed cells.
