E74-like ETS transcription factor 2 (ELF2) is a member of the ETS family of transcription factors, also known as new Ets-related factor (NERF)[1][3][5][7]. In humans, ELF2 is encoded by the ELF2 gene and is expressed as distinct isoforms (notably ELF2A and ELF2B) due to alternative promoter usage[2][4][5][6]. ELF2 isoforms regulate a wide range of genes critical for cell proliferation, cell cycle progression, hematopoietic differentiation, apoptosis, and angiogenesis[2][4][5][6]. ELF2A mainly acts as a transcriptional activator, while ELF2B functions as a repressor and can induce apoptosis and inhibit cell proliferation in certain contexts[2][4][6]. ELF2 plays a prominent role in early B and T cell development. Dysregulation of ELF2 has been linked to oncogenesis and progression of certain cancers, with evidence suggesting roles as both tumor suppressor and promoter depending on isoform and cellular context[2][4][6]. No small molecule or biologic drugs specifically targeting ELF2 or its downstream pathways have been reported, and ELF2 is not commonly classified as a canonical therapeutic target (such as a receptor, enzyme, transporter, etc.)[1][8][7].
