E74 like ETS transcription factor 4 (ELF4)

Target
ELF4
Molecular classification
Transcription factor, ETS family (helix-turn-helix ETS domain-containing protein), Nuclear DNA-binding protein
01

Overview

E74 like ETS transcription factor 4 (ELF4) is a member of the ETS family of transcription factors, characterized by a central DNA-binding ETS domain. ELF4 regulates the transcription of genes vital for hematopoietic homeostasis, immune system development, cell differentiation, and cell cycle progression. It is expressed in normal hematopoietic tissue, ovary, placenta, colon, and certain cancer cell lines. ELF4 functions as both a **tumor suppressor** and an oncogene, depending on cellular context, and plays a critical role in cancer metastasis, proliferation, and immune regulation. Its activity is modulated by post-translational modifications, gene fusions, and cellular signaling networks. ELF4 also participates in pathways controlling apoptosis and DNA damage responses, interacting with proteins such as p53, MDM2, RUNX1, and RUNX2. Clinical research highlights ELF4’s potential as both a biomarker and a target for therapeutic intervention.

Mechanism of action

Drugs targeting ELF4 would likely function by **modulating gene transcription** through ELF4’s DNA-binding activity or by interfering with its protein-protein interactions (e.g., with RUNX1, RUNX2, MDM2). Potential modulations: suppression/activation of transcriptional output in cancer or immune cells. Antagonists of ELF4 could indirectly affect the cell cycle, apoptosis, immune activation, or differentiation.

03

Biological functions

Regulation of gene transcriptionControl of cell cycle, particularly in hematopoietic stem cells and CD8+ T cellsImmune system development, including natural killer cell maturationRegulation of differentiation (osteogenic, adipocyte, neuronal)Apoptosis, especially in response to DNA damageRegulation of interferon production and antiviral immunityModulation of tumor cell proliferation, metastasis, and invasion
04

Disease associations

Cancer (tumor suppressor and oncogenic roles; cancer metastasis, proliferation, poor prognosis in colorectal cancer)Immune dysfunction (deficiency or mutation can cause immune-related diseases)Potential involvement in infection via modulation of antiviral responsesOther: aberrant bone differentiation, stem cell dysfunction
05

Safety considerations

*Not directly known*, as no ELF4-targeted drugs are on the marketPotential concerns would include risk of **immunosuppression**, impairment of normal hematopoiesis, or abnormal differentiation if ELF4 function is excessively modulatedELF4 dysregulation is associated with both enhanced oncogenic proliferation and immune deficiency, representing a therapeutic challenge
06

Interacting drugs

There are *no currently approved drugs* that directly target ELF4; however, it is discussed as a candidate for future drug development and clinical translation

Biomarkers

**ELF4 expression levels**: proposed as a **prognostic biomarker** for cancer progression, especially colorectal cancerIts activity or levels could monitor hematopoietic, immune, or malignant states
