Overview

CD69 molecule is a type II transmembrane C-type lectin domain receptor rapidly expressed on the surface of T-lymphocytes, NK cells, B cells, and platelets following immune activation. It serves as one of the earliest markers of lymphocyte activation and directly regulates cell trafficking by interacting with the sphingosine-1-phosphate receptor 1 (S1P1), retaining activated lymphocytes within lymphoid tissues to support effective immune responses. CD69 modulates diverse intracellular signaling pathways—such as JAK/STAT, mTOR, and ERK1/2—to influence processes like Th17 or regulatory T cell differentiation, immune tolerance, and inflammation. It is implicated in autoimmunity, infection, and as a potential functional target for monoclonal antibody-mediated therapies in immunological research.
