Overview

Early B-cell factor 2 (EBF2) is an atypical helix–loop–helix transcription factor in the COE (Collier/Olf/EBF) family that orchestrates cell fate and gene expression across multiple tissues. EBF2 is a master regulator of brown adipocyte identity, controlling the thermogenic gene program critical for non-shivering thermogenesis in brown and beige adipose tissue, acting partly through modulation of PPARγ and interaction with lncRNAs such as Blnc1. EBF2 is also crucial for proper neural development, where it directs neuronal migration, differentiation, survival, and the establishment of neural circuits—especially in the cerebellum, neocortex, and regions involved with hormone production. In bone, EBF2 participates in the differentiation of osteoclasts by directly regulating genes such as Opg and influences bone mass. Genome-wide association studies implicate EBF2 in predisposition to certain connective tissue disorders, and it has been linked to tumor biology, especially osteosarcoma, where EBF2 expression supports malignant behaviors. EBF2 is thus a non-redundant, pleiotropic transcriptional regulator with critical roles in metabolism, development, and disease, but it is not currently a direct therapeutic target or known drug interaction partner[1][2][3].
