Early growth response protein 4 (EGR4) is a zinc finger transcription factor of the EGR family that regulates gene expression in response to cellular stimuli. It is expressed in various tissues and is implicated in the regulation of cell growth, differentiation, and inflammatory response genes. EGR4 exerts its function primarily by binding to the consensus DNA motif 5'-GCGGGGGCG-3', thereby activating or repressing transcription of downstream genes. In immune cells, EGR4 can interact with NF-κB subunits to strongly enhance the transcription of genes including those critical for cytokine signaling (e.g., IL-2, TNF-α, ICAM-1). EGR4 is associated with proliferative and developmental processes and has emerging roles in certain pathologies, including cancers and reproductive disorders[1][3][4][6].
No approved drugs with direct targeted mechanism; EGR4 acts by DNA sequence-specific activation of target genes and forms complexes with NF-κB components to regulate inflammatory and cytokine gene promoters[1][6].
