Overview

The Eastern equine encephalitis virus (EEEV) envelope glycoprotein is a heterotrimeric complex of E1 and E2 subunits that facilitates viral entry into host cells. The E2 glycoprotein mediates primary attachment to host receptors, including the very low-density lipoprotein receptor (VLDLR), while the E1 glycoprotein is a class II fusion protein that triggers membrane fusion in response to the acidic environment of the endosome (Clark et al., Nature 2022). EEEV is among the most lethal mosquito-borne viruses in North America, causing severe encephalitis with mortality rates up to 70% (CDC, 2024). Because these glycoproteins are the only proteins exposed on the viral surface, they are the primary targets for neutralizing antibodies and vaccine candidates currently in preclinical and clinical development. Therapeutic strategies often involve monoclonal antibodies, such as EEEV-143, which bind to the E2 protein to prevent viral docking and subsequent infection of neuronal cells (Adams et al., 2021). Currently, there are no FDA-approved vaccines or specific antiviral treatments for EEEV infection, highlighting the importance of this target for medical countermeasure development.
