Overview

The Eastern equine encephalitis virus (EEEV) envelope polyprotein is a critical structural component of the EEEV virion, an alphavirus belonging to the Togaviridae family. This polyprotein is post-translationally cleaved into several distinct subunits, primarily E2, which facilitates viral attachment to host cell receptors, and E1, which mediates pH-dependent membrane fusion within the endosome. These glycoproteins form heterodimeric spikes on the viral surface that are essential for the infection of neuronal and myeloid cells. Given its role in the early stages of the viral life cycle, the envelope polyprotein is the primary target for neutralizing antibodies and vaccine development. Current therapeutic strategies focus on monoclonal antibodies that block the E2 receptor-binding domain or prevent the E1 conformational change required for fusion. Understanding the structural biology of this polyprotein is vital for addressing EEEV, which causes a severe and often fatal encephalitis in humans and equines with no currently approved antiviral treatments.
