Overview

Ecotropic viral integration site 2B protein homolog (EVI2B, also known as CD361) is a transmembrane glycoprotein encoded within an intron of the NF1 gene. It is predominantly localized to the plasma membrane and is expressed widely in hematopoietic cells, with highest levels in mature granulocytes. EVI2B is required for granulocyte differentiation and proper function of hematopoietic progenitor cells, influencing their cell cycle progression and survival. It acts as a direct downstream target of the transcription factor C/EBPα. Loss or knockdown of EVI2B impairs myeloid lineage development and hematopoietic stem/progenitor cell functionality. Expression of EVI2B can be dysregulated in acute myeloid leukemia, especially in cases with CEBPA mutations, and may also play a role in modifying the phenotype of neurofibromatosis type 1 and its associated risk of malignancies. The precise molecular function remains under investigation, and EVI2B is not currently targeted by approved therapeutics or drugs
