Ecotropic viral integration site 5-like protein (EVI5L) is a TBC domain–containing GTPase-activating protein (GAP) that modulates Rab GTPase activity, notably Rab10 and Rab23, thereby controlling primary cilium formation and membrane trafficking[1][2][3][4][8]. It is primarily located on intracellular membranes and is involved in negative regulation of ciliary assembly, intracellular protein transport, and positive regulation of GTPase activity[1][4][8]. EVI5L also interacts with several protein complexes, and variants or non-coding RNAs from its locus are associated with several physiological and pathological states such as urolithiasis and X-linked intellectual disability, though direct disease causality in humans is not strongly established[1][2][3].
