Overview

Ectodermal-neural cortex protein 1 (ENC1) is an actin-binding cytoskeletal protein encoded by the ENC1 gene. It contains a BTB/POZ domain and a kelch repeat domain, classifying it within the kelch family of proteins. These structural domains mediate protein-protein interactions and cytoskeletal association[6][2]. ENC1 plays an important role in neural development, differentiation, and cytoskeletal organization, with high expression in the brain and neural tissues[6][4]. Functionally, ENC1 regulates the cellular oxidative stress response, suppresses Nrf2 at the translational level, participates in cell cycle withdrawal during neuronal differentiation, and may be involved in the regulation of apoptosis. It is considered a p53-induced gene, pointing toward its involvement in tumor suppression and cellular responses to stress[1][7][2]. Studies have implicated ENC1 in the progression of several cancers, particularly in neuroblastoma, and its altered expression can serve as a marker of disease. No approved drugs specifically target ENC1, and it is not a therapeutic target or receptor in the conventional sense, but its biological significance in cancer and neural development is notable and may present future therapeutic or diagnostic opportunities[7][4][3].
