Ectonucleoside triphosphate diphosphohydrolase 4 (ENTPD4) is an enzyme of the apyrase protein family that hydrolyzes nucleotide diphosphates and triphosphates in a calcium- or magnesium-dependent manner[1]. Predominantly localized to the Golgi apparatus and possibly lysosomes, ENTPD4 contributes to the recycling of nucleotides during protein N-glycosylation, supporting glycoprotein folding and quality control[1]. The enzyme displays unique features in its nucleotide-binding site and membrane-interacting loops, conferring distinctive substrate selectivity. In cellular stress contexts, particularly during adenovirus infection, ENTPD4 modulates nonclassical apoptotic responses, a role demonstrated by increased cell death when overexpressed in the presence of the adenoviral E4orf4 protein[1]. While polymorphisms have been studied for disease association (e.g., schizophrenia), clear disease linkage is not established, and no direct therapeutic pharmacology is currently reported[1].
