Overview

EF-hand domain-containing protein D2 (EFHD2), also known as Swiprosin-1, is a conserved calcium-binding protein characterized by two EF-hand motifs, a polyalanine region at the N-terminus, and a C-terminal coiled-coil domain[1][2]. EFHD2 is expressed in various tissues, with the highest abundance in the brain, and is involved in processes such as regulation of the actin cytoskeleton, calcium signaling, cell migration, and apoptosis[2][3]. In the immune system, it functions in B-cell receptor signaling as a scaffold protein, influencing calcium flux and survival signaling[1][3]. In neurons, EFHD2 modulates synapse formation and is associated with neuropathological tau protein aggregates implicated in neurodegenerative diseases such as Alzheimer’s disease[3]. Although compelling in cellular biology and disease association, EFHD2 is not currently an established pharmacological target, and no approved drugs are known to directly interact with this protein.
