Eg5, also known as kinesin-5 or KIF11, is a plus-end directed motor protein essential for mitosis. It forms a homotetrameric complex that cross-links anti-parallel microtubules within the mitotic spindle, driving their separation to establish and maintain spindle bipolarity. Due to its critical role in cell division, Eg5 is a target for anticancer therapies, with small-molecule inhibitors being developed to disrupt spindle formation and induce apoptosis in rapidly dividing cancer cells. Emerging evidence suggests it may have non-canonical functions in interphase as well.
