Overview

EH domain-binding protein 1 (EHBP1) is an adaptor protein widely expressed in mammalian tissues, containing a calponin homology (CH) domain for actin binding, an N-terminal C2 domain for membrane association, and a C-terminal bMERB domain for Rab8 family GTPase interaction[1][2][5]. EHBP1 regulates endocytic vesicle trafficking, particularly recycling endosomes and GLUT4-containing vesicles in adipocytes, by linking endosomes to the actin cytoskeleton[1][2][3]. Its molecular function is auto-inhibited until engagement with Rab8 family proteins, which releases the actin-binding CH domain and promotes vesicle movement or membrane tubulation[2]. EHBP1 collaborates with proteins like EHD1, EHD2, and Rab8/Rab10, and disruptions in this protein can affect metabolic processes and have been linked to cancer aggressiveness and development in model organisms[1][2][3][4].
