Overview

EH domain-binding protein 1-like protein 1 (EHBP1L1) is a cytoskeletal adapter protein that contains an N-terminal C2-like domain, a calponin homology (CH) domain, and a C-terminal bivalent Mical/EHBP Rab binding (bMERB) domain, which are conserved in vertebrates. EHBP1L1 acts as an effector for Rab8/10 GTPases and participates in vesicle trafficking by linking vesicular transport to actin dynamics, facilitating transport from the trans-Golgi network to the apical plasma membrane in polarized cells. It is strongly implicated in acrosome biogenesis during spermatogenesis, with specific expression in spermatocytes and spermatids in rodent testes, but is absent from mature spermatozoa. Currently, there is no evidence that EHBP1L1 is a direct target of therapeutic drugs, nor is it classified as a receptor, enzyme, transporter, or traditional drug target class.\n\nSupporting details:\n- Structure: Contains C2-like, calponin homology, and bMERB domains, mediating interactions with actin and Rab8/10.\n- Function: Adapter in vesicle trafficking and cytoskeletal organization, especially important in spermatogenesis and epithelial cell polarity.\n- Clinical relevance: Linked genetically to rare conditions (hydrops fetalis, megaesophagus), but not a direct therapeutic target.\n- Marker: Proposed as a marker for active spermatogenesis in research settings.\n\nNo current drug interactions, biomarker use is primarily experimental, and no established safety or therapeutic concerns are reported in literature or databases.
