Eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) membrane incorporation refers to the physiological process by which these long-chain omega-3 polyunsaturated fatty acids are integrated into the phospholipid bilayer of cellular membranes. This process is a critical determinant of membrane physical properties, such as fluidity and the organization of lipid rafts, which in turn modulates the activity of membrane-bound proteins and signaling pathways (Calder, 2010). By displacing arachidonic acid, EPA and DHA reduce the production of pro-inflammatory eicosanoids and instead serve as precursors for the synthesis of specialized pro-resolving mediators (SPMs) like resolvins and protectins (Mason et al., 2016). Therapeutically, increasing membrane levels of these fatty acids is used to manage hypertriglyceridemia and reduce cardiovascular risk, as evidenced by the Omega-3 Index being a validated biomarker for cardiovascular health (Harris & Von Schacky, 2004). Drugs like icosapent ethyl and various omega-3 supplements work by providing high concentrations of these fatty acids for systemic distribution and subsequent membrane uptake.
