Overview

The Eimeria thiamine transporter is a vital membrane protein found in protozoan parasites of the genus Eimeria, which are the causative agents of coccidiosis in poultry and livestock. As these parasites are unable to synthesize thiamine (vitamin B1) endogenously, they rely entirely on this transporter to scavenge the nutrient from the host's intestinal environment (James, 1980). Thiamine is a crucial cofactor for enzymes in the glycolytic and citric acid cycles, such as pyruvate dehydrogenase, making its uptake essential for the parasite's energy production and rapid replication (Warren, 1968). This transporter is the primary molecular target for amprolium, a structural analog of thiamine used extensively in veterinary medicine. Amprolium acts as a competitive inhibitor, binding to the transporter with high affinity and preventing the entry of thiamine into the parasite cell, which effectively starves the parasite and halts its development during the schizont stage (Merck Veterinary Manual). While the transporter is highly selective for the parasite over the host, prolonged use of inhibitors can potentially interfere with host thiamine absorption, and the emergence of resistant Eimeria strains remains a significant challenge in agricultural disease management.
