Overview

Elastin fiber is a macromolecular structure found abundantly in the dermal connective tissue and is composed predominantly of **elastin**—a durable, extensible, and highly elastic protein encoded by the ELN gene—and microfibrillar proteins such as fibrillin[3][1]. These fibers confer stretchiness and the ability to recoil to the skin and other connective tissues, allowing the skin to return to its original shape after deformation[6][2]. In the dermis, elastin fibers work synergistically with collagen to support mechanical resilience and tissue architecture[1][7]. Elastin fibers are synthesized from tropoelastin precursors that undergo extensive cross-linking, forming mature fiber networks essential for skin elasticity and overall connective tissue integrity[4][5]. Loss, fragmentation, or degradation of dermal elastin fibers is a hallmark of aging, photo-damage, and certain degenerative diseases[6][5]. Elastin fibers are not classic therapeutic targets (like receptors or enzymes) but are important as structural/matrix components in tissue engineering, anti-aging interventions, and some fibrosis-related therapies[6].
