Overview

Elastin microfibril interfacer 2 (EMILIN2) is a glycoprotein component of the extracellular matrix (ECM), belonging to the Emilin/Multimerin family, and is widely expressed in tissues such as bone marrow, skin, and the cochlea[1][3][2]. In bone marrow, EMILIN2 forms part of the non-cellular matrix microenvironment, regulates mesenchymal stem cell differentiation (notably inhibiting adipogenesis), and impacts hematopoietic progenitor and stem cell frequencies, especially during aging. Loss of EMILIN2 impairs normal bone marrow function and stem cell differentiation, and its expression is deregulated in certain cancers, especially acute myeloid leukemia, suggesting a possible role in malignancy development and as a disease biomarker[1]. In other tissues, such as the cochlea, EMILIN2 helps to organize extracellular architecture and impart mechanical properties important for auditory processing, while in skin it is expressed mainly in the microvascular endothelium, implicating EMILIN2 in regulation of vascularization and interacting with pathways such as EGFR-dependent signaling for angiogenic processes[2][3]. No approved drugs directly target EMILIN2, and it is not considered a therapeutic target such as a receptor or enzyme but rather a structural/modulatory ECM protein with regulatory functions.
