Overview

Elastin microfibril interfacer 3 (EMILIN3) is an extracellular matrix glycoprotein in the EMILIN/Multimerin protein family, characterized by an N-terminal cysteine-rich EMI domain. EMILIN3 is the least characterized member of its family and displays a dynamic and tissue-specific expression pattern, mainly localizing in developing gonads, osteogenic mesenchyme, and connective tissues. In skin, it localizes to the tips of oxytalan fibers and specialized basement membranes surrounding hair follicles and sebaceous glands. It serves structural roles in the extracellular matrix, can oligomerize, and modulates signaling by functioning as an extracellular antagonist of transforming growth factor β (TGF-β) ligands. EMILIN3 is reported as a marker of changes in the dermal extracellular matrix and as potentially involved in carcinogenesis and tissue remodeling, although it is not described as a direct therapeutic target and there are no known pharmacological agents directed against it.
