Electrophilic metabolites of 4-hydroxyalkenals (such as 4-hydroxynonenal or 4-HNE) are highly reactive α,β-unsaturated aldehydes generated during lipid peroxidation, particularly following oxidative degradation of polyunsaturated fatty acids. Similarly, reactive oxygen species (ROS) are chemically reactive molecules derived from oxygen. Both can react with nucleophilic residues in proteins, DNA, and other biomolecules, leading to cellular damage, disruption of cellular signaling, and the initiation of detoxification responses such as glutathione conjugation. Though vital in small amounts for redox signaling, elevated levels are associated with oxidative stress and contribute to the pathology of cancer, neurodegeneration, inflammation, and cardiovascular diseases. These entities are not specific therapeutic targets (like receptors or enzymes) but represent classes of reactive intermediates that challenge cell survival and homeostasis.
