Overview

ENL, also known as MLLT1, is a nuclear protein that plays a crucial role in chromatin regulation and transcriptional elongation. It functions as a histone acetylation reader, recognizing acetylated lysine residues on histone H3 via its YEATS domain. ENL is also a component of the super elongation complex (SEC), facilitating RNA polymerase II-mediated transcription. Aberrant ENL activity, particularly through MLL–ENL fusion in acute leukemias, drives oncogenic gene expression. The YEATS domain is a therapeutic target, with small-molecule inhibitors under investigation for treating AML.
