Overview

ELF3 antisense RNA 1 (ELF3-AS1) is a human long non-coding RNA (lncRNA) transcribed from the antisense strand of the ELF3 locus. It does not encode a protein but acts as a regulatory RNA implicated in oncogenic processes. ELF3-AS1 is upregulated in several cancers, including osteosarcoma and bladder cancer, where it promotes tumor cell proliferation and disease progression. Mechanistically, ELF3-AS1 upregulates the oncogenic transcription factor KLF12 by promoting methylation and downregulation of the tumor suppressor miR-205. Its aberrant expression is correlated with higher cancer stage and more aggressive phenotype, supporting its candidacy as a potential diagnostic biomarker and experimental therapeutic target in oncology.

Mechanism of action

Not applicable for drugs, but mechanistically, ELF3-AS1 appears to promote cancer cell proliferation by: Upregulating Kruppel-like factor 12 (KLF12) via downregulation/methylation of miR-205; Acting as a competitive endogenous RNA or through epigenetic modification

03

Biological functions

Regulation of gene expression at the epigenetic and post-transcriptional levelRegulation of cell proliferationPotential modulation of DNA methylation (specifically, methylation of microRNA genes such as miR-205)Oncogenic activity in specific cancers such as osteosarcoma and bladder cancer
04

Disease associations

Cancer (implicated in osteosarcoma, bladder cancer, and potentially other malignancies)Other (investigated in the context of cell cycle progression)
05

Safety considerations

Not established, as there are no drugs clinically targeting ELF3-AS1 currently; potential therapeutic targeting may have off-target or epigenetic toxicity concerns (speculative, based on lncRNA function).
06

Biomarkers

Overexpression of ELF3-AS1 may serve as a biomarker for: Poor prognosis or aggressive disease in osteosarcomaProgression in bladder cancer
