Overview

ELMO domain-containing protein 1 (ELMOD1) is a member of the ELMOD family of proteins, which are ancient and highly conserved through eukaryotes. ELMOD1 functions as a GTPase-activating protein (GAP) specifically for ARF and ARL family small GTPases. It plays essential roles in the regulation of ciliogenesis, protein import into cilia, and protein trafficking from the Golgi to cilia. Mouse knockout studies indicate that ELMOD1 is important for the formation and maintenance of primary cilia and hair cell stereocilia, with loss-of-function resulting in hearing and balance deficits due to compromised actin-based hair cell bundle development. Unlike canonical drug targets such as receptors or enzymes that are directly targeted by current therapeutic drugs, ELMOD1 is best categorized as a cell signaling/regulatory protein with no established pharmacological modulators or clinical drug interactions at this time[1][2][3].
