Overview

Elongation factor 2 (EF-2) is a eukaryotic GTP-binding translation elongation factor that catalyzes the translocation step during protein synthesis, moving the mRNA and ribosome after peptide bond formation. It is an essential component of the protein synthesis machinery. Pseudomonas exotoxin A specifically inactivates EF-2 through ADP-ribosylation, transferring the ADP-ribose moiety from NAD+ to a histidine residue (diphthamide) in EF-2, thereby blocking translation and causing cell death. This mechanism is similar to that of diphtheria toxin.
