Overview

Elongation factor for RNA polymerase II 3 (ELL3) is a protein-coding human gene that encodes a transcription elongation factor within the ELL family[2][9][10]. ELL3 acts by promoting transcription elongation by RNA polymerase II, principally by suppressing transient pausing of the polymerase during mRNA synthesis[4][2][10]. ELL3 exhibits context-dependent functions, being especially important in gene regulatory events in embryonic stem cells, including enhancer priming and the regulation of genes crucial for cell identity and differentiation[2][10]. ELL3 has also been implicated in oncogenic processes, cell fate determination, and has a potential (but as yet not established) connection to diseases such as cancer, though concrete evidence for direct clinical targeting or associated drugs is not available at this time[2][10][6]. ELL3 shares homology and some overlapping functions with other ELL family members but displays tissue-specific expression patterns (notably more restricted to testis in mammals)[4][8][10]. It is a central component of the transcription elongation complex, with functional domains mediating both positive and regulatory influences on RNA polymerase II activity[2][1][7]. There are currently no known drugs that specifically target ELL3, nor are there established biomarkers or notable safety concerns related to modulation of this factor in clinical settings.
